Abstract
The effect of chronic tricyclic antidepressant (TA) treatment on noradrenergic and serotonergic neurones was studied. Methods Rats (male, Sprague Dawley, 250-300 g) were anesthetized with chloral hydrate (400 mg/kg, i.p.). Electrophysiological recordings were made, by a previously described single-cell recording technique, from the dorsal raphe-nucleus (DRN) or the locus-coeruleus (LC) with or without iontophoretic administration of clonidine (C). Recording sites were verified histologically Animale were pretreated with test drugs. Results Chronic pretreatment with imipramine (I, 10 mg/kg, i.p., 12-hrly for l1-14 days) significantly inhibited the action of C (0.01M) on the firing rate of LC neurones. Yohimbine (1.5 mg/kg, i.v.) but not propranolol (10 mg/kg, i.p.), reversed the action of I. The LC-inhibitory action of C was restored by 72 hr after discontinuation of I. C resistance was also produced by treatment with I (50 mg/kg, i.p. 12-hourly) for 3 days. Chlorimipramine (5 mg/kg,b.i.d. for 14 days) did not alter receptor responsiveness. I pre-treatment did not alter the firing of neurones in the DRN or the effect of LSD (8 and 12 mcg/kg, i.v.).
