Abstract
The lack of valid behavioral animal models represents an intrinsic difficulty for the investigation of the still largely unknown mechanisms of action that subserve human hallucinosis. An experimental approach commonly used is the comparison of effects elicited by psychotomimetic compounds with those induced by chemically related nonpsychotomimetic drugs on brain monoaminergic neurons. Following this line, we have recently investigated d-lysergic acid diethylamide (LSD) and lisuride, a semisynthetic ergot derivative stereochemically related to isolysergic acid which has been claimed to be nonhallucinogenic (16). We determined the effects on the 5-hydroxytryptamine (5-HT), dopamine (DA), and norepinephrine (NE) systems in the brains of experimental animals, as assessed by behavioral, neurochemical, and electrophysiological parameters. The present chapter reviews the main findings on this topic obtained in our laboratories as well as recent pertinent data in the literature.
