“Stimulus Properties of Phenethylamine Hallucinogens and Lysergic Acid Diethylamide: The Role of 5-Hydroxtryptamine”.

Abstract
Experiments were carried out in rats, to examine the antagonism of the stimulus effects of mescaline (M) by BC-105 and cinanserin (CN). The effect on M -mediated stimulus control of 5HT depletion by p-chlorphenylalanine (CPA), an inhibitor of 5HT synthetic, was also studied. Rats were trained using electric shock and made to acquire discrimination between the effects of M-HCl (10 mg/kg, groups 1 and 2 ) or d-amphetamine SO4 (0.8 mg/kg, groups 3 and 4) and saline. The drugs were injected i.p. 60 min before testing. Tests in which the drugs to be antagonized were LSD, 2,5- dimethoxy-4-methylamphetamine (DOM) and 2,5-dimethoxy-4- ethylamphetamine (DOET) were also carried out. Results Pre-treatment of group 1 with increasing doses of BC-105 with M led to a progressive decline in response rate until at 3 mg/kg, responding was 130f the control rate. Rates of responding in group 2 increased as a function of the dose of BC105. The data from groups 1 and 2 indicated a complete and specific antagonism by BC-105 of the stimulus effects of M Similar results were obtained with CN. Maximum antagonism it group 1 occurred at 10 mg/kg while in group 2 it occurred at 3 mg/kg of CN. In group 3, pretreatment with 10 mg/kg of BC-105 produced a modest decline in responding and, at 30 mg/kg, the response rate was indistinguishable from the saline condition. Neither 10 nor 30 mg/kg significantly changed the responding in group 4. Cross tests with LSD (50 mcg/kg) DOM (0.3 mg/kg) and DOE (0.3 mg/kg) yielded response rates appropriate for the M condition. Pretreatment with the maximally effective doses of BC-105 or CN, blocked the M-like effects of LSD and the phenylalanines. Pretreatment with CPA had no effect onM as a discriminate stimulus. Drugs which mimic the stimulus effects of M do so via a common, serotoninergic mechanism.
