Abstract
The analysis of Ergot alkaloids by HPLC is reported. Ergotamine, ergocristine and dibydroergotamine were obtained by plant extraction. Ergocornine, ergocryptine and ergometrine were isolated from a fermentation process. The dihydroderivatives listed below were obtained from both sources. The extracts were applied to LiChromosorb or Silica SI or LiChrosorb or Silica RP-8 columns with CHCl3-EtOH (95.5), CHCl3-n-hexane-EtOH (40:40:10) and CHCl3-MeOH as eluants. The compounds listed below were isolated. The internal standards 1-phenyl-2,3-dimethyl-4-dimethylamino-5-pyrazolone- and 2propylthiocarbamoyl-4-pyridine were for ergo/amine-containing samples and ergocristine-containing samples respectively. The individual ergotoxine alkaloids listed below were separated on reversed-phase columns using bonded-phase packings containing C2, C8 and C18 alkyl groups. A high-efficiency microparticulate reversed-phase column was used to separate the active compounds as stereo and structural isomers.
