“LSD and Tryptamine Effects on Sleep/Wakefulness and Electrocorticogram Patterns in Intact Cats”.

Abstract
The effects of LSD and tryptamine (TR) on sleep/waking and EEG patterns were studied in cats. Methods Sleep/waking patterns were measured in 6 male cats (3.94.5 kg) by monitoring 2 electrocorticograms (ECG)(visual and others), a bilateral electrooculogram, a nuchal electromyogram and a hippocampal EEG. The effects of a 5 min i.v. infusion of 3.75, 7.5 or 15.0 ug/kg LSD, or 150 min infusions of 0.04, 0.08, or 0.12 mg/kg/min TR or -saline were determined. Results LSD Produced a dose-related increase in wakefulness and decreases in REM and spindle sleep for the 1st 75 mins without affecting EEG frequencies. During the 2nd 75 min period the increase in active wakefulness and decrease in REM sleep continued but spindle sleep increased. Increases in delta index and ECoG amplitude with a decreased frequency were significant in period 2. TR Caused a dose-related increase in wakefulness during the 1st 75 min of infusion with significant decreases in spindle and REM sleep throughout the infusion period. Changes in ECoG were small, amplitude increased, delta index tended to decrease and faster ECoG frequencies were more prominent. Effects disappeared at the end of the infusion. Injection of 10 lug TR into the 3rd ventricle suppressed REM sleep within 2 min and caused active wakefulness. ECoG Spindling returned at 34 and 53 min but was abolished by a 2nd injection. Conclusions Both LSD and tryptamine increase wakefulness and decrease REM and spindle sleep. The mechanism of the arousal effect is not yet clear.
