Abstract
The discriminative stimulus properties of cyclazacine (C). pentazocine (A), and nalorphine (N) were compared in rats. Method s Rats (about 6-wk-old, mean weight 416 g) were trained to discriminate P (Sterling-Winthrop, 10 mg/kg s.c.), C (SterlingWinthrop, 0.5 rag/kg s.c.), and N (20 mg/kg s.c.) from saline. After injection of the assigned drug, depression of 1 lever resulted in food reinforcement; the opposite lever was reinforced after saline. Stimulus generalization tests to each other narcotic antagonist, d-amphetamine (0.1-1.6 :ng/kg 8.C.), morphine (0.62.5 mg/kg s.c.), and LSD (0.025-0.4 mg/kg s.c.) were performed. The effect of naloxone (Undo, 0.4-3.2 mg/kg s.c.) on the discrimination of P. C and N was determined. Results Discriminated responding was readily acquired with all 3 drugs, and the accuracy of discrimination was related to drug dose. Naloxone antagonized discrimination of C, P and N. Stimulus generalization tests indicated that C, P and N each had highly specific stimulus properties. Clear stimulus generalizations occurred only to P and C in the N-saline group, but neither C nor P generalized to N.
