“Comparison of the Action of Lysergic Acid Diethylamide and Apomorphine on the Copulatory Response in the Female Rat”.

The effects of LSD and apomorphine (AM) on copulatory behavior were studied. Methods SPF Sprague Dawley female rats (250-300 g) were ovariectomized. Before drug testing the rats were tested with estradiol benzoate (Organon; 10 mcg/kg) followed 48 hr later by progesterone (Organon, 0.4 ma) for an assessment of lordosis frequency in comparison with the laboratory standard. - Drug effects were evaluated 2 wk later. A predrug test was done to ensure that the hormone treatment had taken effect. Both chlorpromazine (CPZ; Hibernal, Leo, ampules 25 mg/ml) and pimozide (PZ; Janssen) were injected 1 hr before LSD and AP (both Sandoz). The females were tested individually in the home cage of stud Wistar males, selected for sexual vigor, where a maximum of 6 mounts was allowed. The 0.000000emales showing lordosis in a given treatment group was calculated from D total response; 2 consecutive lordoses out of a total of 6 mounts and from 2) early response: 2 consecutive lordoses already in the first 3 mounts. Locomotor ability was tested for 10 min using anAnimex activity meter. Results Both LSD and AP dose-dependently inhibited lordosis behavior and showed their maximal inhibitory effects on lordosis at the first test, 10 min after the injection. Recovery from this effect had come menced at the following test 30 and 50 min later, respectively. PZ prevented lordosis inhibition by AP (0.2 mg/kg) partially at 0.1 mg/kg and completely at 0.5 mg/kg, whereas it had no effect on LSD-induced inhibition. CPZ at 1.0 mg/kg prevented the lordosis response but had no effect at 0.5 mg/kg. When combined with LSD, PZ at 0.5 mg/kg had no effect in preventing the lordosis inhibition by LSD. There were no significant effects obtained on the APinduced lordosis irdlibitor by CPZ. All treatments impaired performance on the locomotor tests. The predominant action of LSD on the female copulatory response is not mediated by increased dopamine receptor activity but the LSD effect may be modified by decreased dopaminergic activity.
