Abstract
Biliary excretion of (3H)lysergic acid diethylamide (LSD) was compared in normal Wistar and the homozygous Gunn rat in which jaundice persists through life. Methods - Male Wistar and Gunn rats (250-350 g) were fitted with femoral vein, carotid artery and biliary catheters under urethane anesthesia (10 ml of 14%kg; i.p.) (3H)LSD (24 microCi; 0.5 mcg/kg; i.v.) (Amersham) was injected. Blood and bile samples were taken over 150 min. examined for radioactivity and examined by TLC before and after -glucuronidase incubation. In the Wistar rat 460f the dose of (3H)LSD was recovered in the bile in 2.5 hr. The main metabolites were identified as glucuronides (M3 and M4), their hydrolysis by 13 - glucuronidase being inhibited by glucaro-(1 .4)-lactone (5 mM) (Calbiochem). In the Gunn rat only 260f the dose of (3H)LSD was recovered in the bile in 2.5 hr but the metabolic pattern was identical to that in Wistar rats. Conclusions Despite the genetic lesion in UDP-glucuronyl transferase in the Gunn rat, the major part of LSD is still excreted as glucuronides, The enzyme may therefore be present at a low level or the Gunn rat may be deficient in only some of a number of enzymes. Edrophonium is also excreted aS a glucuronide in the Gunn rat.
