Abstract
The effect of electroconvulsive shock (ECS) on cerebrocortical PG content was studied. Brains were removed from male Sprague-Dawley rats (200 g) and the cerebral cortices frozen homogenized and PG's extracted, separated by chromatography and estimated by radioimmunoassay. The cortical level of PGF was 99.1+/-13.5 ng/g and of PGE, ll+/- 7.5 ng/g. Amphetamine (10 mg/kg i.p.), chlorimipramine (20 mg/ kg i.p.), phenoxybenzamine (25 mg/kg i.p.) chlorpromazine (10-mg/ kg i.p.), haloperidol (1 mg/kg i.p.) and LSD (0.2 mg/kg i.p.) had no effect on PG levels. Pentylenetetrazole -(50 mg/kg i.p.) markedly increased levels of PGF's within 2 min. reaching a peak in 5 min and returning to normal witbin 30 min. Indomethacin (10 mg/kg i.v.,Merck-USA) reduced PGF levels and pretreatment with 20 mg/ kg i.p. prevented the increase caused by ECS, but did not block the convulsions.
