“Noradrenergic Regulation of Brain Serotonergic Neurons: Evidence from Single Unit Studies with Clonidine”.

Abstract
Previous studies show that the antihypertensive agent clonidine inhibits noradrenaline (NA) release from peripheral and central adrenergic neurons. The drug also, in mcg-doses, decreases the turnover rate of brain 5-hydroxytryptamine, (5-HT). The effects of clonidine on the firing rates of brain NA neurons were investigated in n. locus coeruleus and serotonergic (5-HT) neurons in n. raphe dorsalis by means of single-cell recording. NA neurons and a majority of 5-HT neurons were inhibited by cionidine (mean dose of 5.7 and 9.0 mcg/kg i.v., respectively). The effect on the 5-HT neurons seemed to be secondary to'the effect on the NA neurons since destruction of the NA input by pretreatment with intraventricularly administered 6 -hydroxydopamine (100 mcg, I wk) abolished the effect of clonidine on the raphe. In contrast, this pretreatment did not antagonize LSD, which inhibits the raphe neurons directly. Also, the NA-releasing drugs, d- or l-amphetamine (0. 5mg/kg i;v.), reversed the effect of clonidine on the raphe. The results support the hypothesis that brain NA neurons may exert a regulatory influence on 5-HT neurons in the raphe .
