“LSD as an agonist dopamine receptors in the striatum”.

Abstract
Possible activation of dopamine in corpus striatum by (+)-lysergic acid diethylamide (LSD) was investigated. Method The rotational responses of male SPF rats with unilateral damage to their nigro-striatal system (induced by the injection of 10 mcg 5,6 dihydroxytryptamine (5,6-HT) or 3.5 mcg 6-hydroxydopamine (6-OH - dopamine) into the right medial forebrain bundle) to 1-5 mg/kg of (+)-methamphetamine, 0.05-3 mg/kg apomorphine or 100,200 or 1,500 mcg LSD/kg i.p. were determined. Results 5,6-HT and 6-OH-dopamine produced similar depletion of dopamine stores in the telediencephalon on the ipsilateral side of injection. 6-OH-dopamine also lowered noradrenaline contents without affecting 5-HT (5-hydroxytryptamine) levels. 5,6-HT lowered 5-HT without changing noradrenaline contents.- (+) - Methamphetamine produced ipsilateral and apomorphine contralateral circling in rats pretreated with either of the neurotoxics. :Control rats showed no such rotational responses to amphetamine or apomorphine. The 100 mcg/kg dose of LSD tartrate, but not the lowest dosage, led to contralateral rotation (i.e. towards the non-operated side) from the 5th min after the injection, reaching maximum after 15 min. The rotational responses did not decrease after repeated applications of 200 mcg LSD/kg on 5consecutive days, thus indicating no induction of tolerance. The rotational responses to LS13 were significantly decreased by 1 mg haloperidol/kg injected i.p. 10 min after the 200 mcg/kg dose of LSD, both in 6-OH-dopamine and 5,6-HT pretreated rats. 200 mg c-Methyl-L-tyrosine/kg injected i.p. 4 hr before LSD, induced sedation but did not affect circling. No rotational responses were induced by up to 20 mg (+)-2-bromo - lysergic acid diethylamide bitartrate (BOL-148)- in the 5,6-HT - or the 6-OH-dopamine-treated rats.
