Abstract
The effects of haloperidol or haloperidol plus LSD on neurons of cortical motor regions in rats were studied. 90 Male white rats aged 3-4 mth were used. The brain was excised 1,4,8,12 and 24 hr after a single dose of 1 mg/kg haloperido, and after LSD (LSD-25) 40 mcg/kg plus haloperidol 1 mg/kg the haloperidol being given 15 min after LSD. Haloperidol had a motor inhibitory effect 2-2.5 hr after administration. In pathomorphological investigation 1 hr after administration gangliose cells showed signs of moderate swelling. After 4 hr. tinctural properties of basic neuron masses had significantly increased, and there were individual hyperchrome neurons with signs of swelling. After 8,12 and 24 hr there was normalization of the condition of gangliose cells and in the cortex in all layers, especially III-V, large hyperchrome neurons were apparent. Combined administration of LSD and haloperidol led to restriction of the period of motor stimulation to 20-30 min. after which the animals became semi-somnolent. 1-4 Hr after administration there were signs of acute swelling of neurons, and in the cytoplasm of neurons, there were increased numbers of free ribosomes and polysomes. Granular endoplasmatic reticulum was vacuolized and the mitochondria much swollen. In later stages, there was a tendency to normalization of ultrastructural organization of neurons. It is suggested that haloperidol has a stimulating effect on the structures of the mitochondria, and this effect is preserved when it is used with LSD.
