“Timid Singly-Housed Mice: Their Value in Prediction of Psychotropic Activity of Drugs”.

Abstract
The selectivity of the timidity-reducing effect of drugs in timid singly-housed mice was studied. - Male mice housed singly for 3-5 wk were studied for interaction with group-housed mice on a 1:1 basis for 4 min. Drugs were administered 30 min before interaction. Incidence of sociable, timid, aggressive and locomotion activities was noted. Rota-rod tests were also carried out. 600f isolates did not attack their partners; 45% appeared 'timid¹, 15% 'sociable'. Isolation-induced timidity was stable in repeated interactions. 5 mg/kg Diazepam p.o. and chlordiazop-oxide (20 or 50 mg/kg) and 60 mg/kg barbitone lowered timidity and increased locomotion and social behavior. 60 mg/kg Barbitone also stimulated aggressive behavior; 20 mg/kg barbitone increased only sniffing and following of partners. 7.5 mg/kg Chlorpromazine inhibited timidity and appeared to have a general depressant effect. 80 mg/kg Imipramine reduced timidity, locomotion and rearing. Dexamphetamine increased alert postures at 0.25 mg/ kg and reduced social behavior; LSD (0.01 and 1 mg/kg) stimulated tail-rattling. In the rotarod tests, lower doses of drugs were needed to induce falling off in singly-housed than in group-housed mice but fewer isclated mice were able to stay on in control tests, 9/25 in contrast to 19/24 group-housed. Recording of sociable behavior in timid mice may help to differentiate effects of sedatives relieving anxiety from those of neuroleptics.
