Abstract
The parikarya of 5-hydroxytryptamine (5HT) containing neurons in the braín are found in the raphe nuclei (Dahlstrom and Fuxe, Acta. Physiol. Scand. 62: 1, 1965). These experiments sought to (1) determine whether electrical stimulation of the dorsal raphe nucleus was a discriminative stimulus and (2) investigate the interáction of some drugs with this stimulus. In 10 rats, bipolar electrodes were implanted in the dorsal raphe nuclei. Following recovery from surgery, the rats were trained to press both bars of an operant chamber and discrimination training was begun. Depression of one of the levers resulted in food reinforcement and presence of brain stimulation (biphasic rectangular pulse pairs, 300-600 microA) and responses on the opposite lever were food reinforced without brain stimulation. A high degree of discriminated responding rapidly occurred. Morphine sulphate 2.5-10.0 mg/kg or LSD tartrate 25-100 mcg/kg did not alter responding during stimulation. Without stimulation, LSD, but not morphine, produced responses appropriate to electrical stimulation. Results indicate that electrical stimulation of the dorsal raphe nucleus and LSD have similar stimulus properties.
