Abstract
D-LSD binding to brain homogenates was studied. Brains were isÚlated, from male Sprague-Dawley rats (7-12 wk old). They were - homogenized and incubated with d-lysergic - acid diethylamide at 0--32 x -1M or with d - LSD and 5-HT (O.1-1.O mM). A rapid filration technique was used to determine high - affinity d-LSD binding in brain fractions. When 5-HT and LSD were incubated together, d-LSD -binding wŠs reduced. Both 'specific' (4 x 10-9M half saturation) and ≥non-specific≤ LSD binding were demonstrated. Specific d-LSD binding occurrŤd in corticŗl and sub-cortical (e.g. striatum and diencephalon) regions,but not in the cerebellum. 5 Min after -d-LSD 7.5 mcg/kg was injected i.v., d-LSD concentrŠtion-in the midbrain was 2-x lO-8M. Specific d-LSD binding in forebrain homogenates was unaltered in-rats with lesions in the raphe nuclei.- Specific d--LSD binding was redýced after incubation with methysergide N,N - džmethyltryptamine, prornethazine-and chlorpromazine, chiorimipramine. imipramine, desimipramine,-cyproheptadine, dl-DOM, mescaline, reserpine, 1-LSD and dopamine. GABA, norepinephrine, glutaminej histamine, carbachol, iproniazid and pargyline had no effect at 0.1-mM. Most of the d-LSD- binding sites may be associated with a post-synaptic serotonin receptor, instead of presynaptic recepors with serotonergic inputs.
