Abstract
The effects of LSD on the biochemical pathway of 5-hydroxy-tryptamine (5-HT) in the brain of growing mice we're investigated. Methods Mice at 1 and 7 days of age and maintained on a lighting regime of 10 hr of light and 14 hr of darkness received single LSD doses of 0.5 mcg/kg, 5.0 mcg/kg and 50mcg/kg i.p. respectively. They were decapitated at 8 wk of age and cerebellum, cerebral hemispheres, mesencephalon-diencephalon and medulla pans assayed for each pathway enzyme, using 9 - l0 animals for each dose. Tryptophan-5-hydroxylase (T5-H) was assayed by modification of the CO2 trapping method with 4C-tryptophan as a substrate. 5-Hydroxytryptophan decarboxylase (5-HTD) was assayed by measuring 5-HT formed from t4C 5 - hydroxytryptophan as a substrate. MAO was assayed by measuring 5-hydroxyindole acetic acid (5-HIAA) formed from 14C 5-HT substrate. Results Injection of LSD to 1- or 7- day-old mice had no effect on T5-FI, 5-HTD or 5-HIAA, but caused augmentation of 5-HT in the mesencephalon-diencephalon of 7-day-old mice at 5.0 mcg and 50 mcg/kg dosage. MAO was elevated in the hemispheres of 7-day-old mice at 5.0 mcg and 50 mcg/kg and in the medumcg pans at 5.0 mcg/ kg. Conclusions None of the changes brought about by LSD exceeded 10%, suggesting that only part of the 5-hydroxytryptamine system could have been modified by the drug. The effects at 7 days of age correlate with the period of rapid terminal formation of brain 5-HT neurons.
