Abstract
To investigate the effect of varying monoaminergic activity levels on motivation, a serotonin agonist (LSD), a dopamine agonist (apomorphine, APO) and MAO inhibitors (p - chlorophenylalanine PGMCG and u-methyl-p-tyrosine, MT) were used. The influence.of these compounds on the urge to drink water in water deprived animals or to seek heterosexual contact in castrated estrogen treated female rats was evaluated. Motivation was determined using the-increasing barrier technique (Meyerson, B.J. and Lindstrom L., Acta Physiol. suppl. 389, 1973.) i.e. the willingness to cross an electrical grid to attain water or a sexually active male. In parallel the effect on spontaneous motor activity (SMA) of the various treatments was recorded. LSD, 0.05mg/kg but not-0.01 mg/kg significantly lowered the number of crossings when the goal-cage held a male. The higher dose clearly depressed SMA. Apomophine (0.05 or O.OZ mg/kg) lowered the number of crossings for water. Identical doses of APO significantly lowered SMA. aMT, 100 mg/kg lowered crossings to reach a male but slightly enhanced crossings for water. PCPA was given for 4 consecutive days (100+50x3 mg/kg) and this enhanced grid cross-ings by females reaching for the male; this effect was found only in estradiol benzoate treated subjects. The same treatment lowered crossings in water deprived animals seeking water.
