Abstract
The psychotomimetic potency of 13 congeners of LSD was determined in normal healthy human beings, all of whom were familiar with the subjective effects of LSD. . The only alteration in the LSD molecule which did not reduce psychotomimetic potency was substitution of an acetyl group on the indole nitrogen (ALD-52). Inactivation was greatest in the case of BOL-148 and MBL-61 in which a bromine atom replaces hydrogen at position two of the indole ring. In contract to previous reports in the literature, BOL-148 consistently caused mild mental changes but did not elicit a complete spectrum of LSD effects when given in doses greater than 70 mcg/kg orally. . High potency as an antagonist of 5-HT in isolated smooth muscle preparations was not correlated with high potency as a psychotomimetic. Thus, MBL-61 exerted no psychotomimetic effect but is reported to be five times as potent as LSD in blocking 5-HT on the isolated rat uterus. . These data do not support but do not disprove the hypothesis that LSD causes a psychosis by competing with 5-HT.
