Abstract
5-hydroxytryptamine (5-HT) given in two s.c. doses of 15 mg/kg produces ulcerations in the stomach and cecum of rats. The ulcerogenic effect is due not to an increase in gastric secretion but to an increase in muscle tone and, as histological study showed, to a vascular effect (hyperemia with isolated ischemia, petechia and edema). . Two s.c. doses of 0.2 or 0.5 mg/kg LSD markedly inhibited this effect of 5-hydroxytryptamine on the stomach and intestine. . 5-hydroxytryptophane, the precursor of 5-HT, had a considerably slighter ulcerogenic effect, even when 2 doses of 300 mg/kg were given s.c. LSD (2 doses of 0.5 mg/kg s.c.) modified this effect also. . Various isomers of 5-HT and its precursor exerted no ulcerogenic effect. Of these isomers, only 5- hydroxyisotryptamine significantly antagonized the ulcerogenic effect of 5-HT. It also inhibited the vasospasm induced in the isolated rabbit ear by 5-HT. It seemed to potentiate the ulcerogenic effect of the precursor of 5-HT.
