Abstract
The response of single raphe neurons to (+)-LSD was evaluated and correlated with binding of the drug in the brain. Male albino rats (200-220 g) were anesthetized with chloral hydrate (400 mg/kg), and the characteristic slow rate of the raphe units monitored electronically following insertion of micropipettes into the brain. After establishing a steady baseline rate for firing of raphe neurons, 3H-(+)-LSD was injected into a tail vein. Raphe firing was rapidly decreased by a dose of 3 mcg/kg, and recovery was slow. Rats were decapitated at various stages of the recovery phase, and the amount of labeled (+)-LSD bound in various parts of the brain was determined. The inhibition of firing of the raphe neurons by (+)-LSD occurs in a concentration range corresponding to high-affinity binding of this drug in vitro. (+)-LSD is retained more effectively in the brain areas rich in LSD-binding sites, e.g. striatum > cortex > hippocampus>diencephalon>midbrain> cerebellum.
