“Lysergic acid diethylamide (LSD-25) antagonists. II. Development of tolerance in man to LSD-25 by prior administration of MLD-41 (1-methyl-d-lysergic acid diethylamide).”.
Arch. Neurol. & Psychiatr.. 1958;79:201.
Abstract
Effect of MLD-41 (1-methyl-d-lysergic acid diethylamide) on Siamese fighting fish was similar to that of LSD but MLD-41 is only about 1/10 as active. . Effect of MLD-41 in normal human subjects was similar to that of LSD (the five subjects could not distinguish between the two drugs). The threshold dose was about 70 mcg orally. . Block by MLD-41 of LSD effect: When MLD-41 was given for 5-6 days before the administraiton of LSD, the effect of the latter was blocked, i.e. tolerance to LSD developed. A total amount of about 1000 mcg MLD-41 blocked the effect of 100 mcg LSD orally. . Comparison of blocking effect of MLD-41 and BOL-148: The production of tolerance to LSD by MLD-41 was about 3-4 times as effective as that by BOL-148. It would seem that the closer the molecule is to LSD in its effect on man and fish, the greater the probability of tolerance production.
